ListMath Description

To Process list files on a PC Becton Dickinson brought PC-LYSES. This program is capable of displaying one and two dimensional parameter plots (i.e. histograms and dot plots/contour plots). It can do gating on all parameters and give al kinds of statistics on them. What PC-LYSES can't do is compensation and display density plots and three dimensional dot plots. We developed a program to process up to 4 parameters from a flow cytometer list file with compensation, thresholding and gating. It gives the possibility to do free calculations with these gates. The program can display parameters as histograms, two dimensional density plots and three dimensional dot plots. The ListMath application can be used to calculate the Tpot from cells labelled with IdUrd as first label, ClUrd as second label and a DNA-label. The program accepts standard flow cytometer list files with at least three and not more then eight parameters. Two, Three or Four parameters must be selected for the X, Y, Z and W axis. It's possible to load fewer events the there are in the list file. Because the more events loaded the slower the program works this is useful for setting compensation and selections. It is possible to change the number of events anytime. After the list file is loaded the six compensation factors can be set. This is done by typing in the value's or setting a compensation line in a density plot. The threshold can be typed in for all three parameters. These thresholds work on the not compensated data. This means that the thresholding on the data is done before the data is compensated. It is possible to set the data in the last channels to zero. These channels normally collect all events outside the measurement window. The X-axis can be separated in 5 regions by setting 4 selection line's (numbered x1s,x2,x3s,x4s) in a density plot, both Y, Z and W-axis can be separated in 4 regions by setting 3 selection line's in a density plot (y1s,y2s,z1s,z2s).
